There are two missing letters that are not used nowadays. One looks like a Y but with three arms at the top pronounced with a 'th' sound - usually it is replaced with a normal Y, as in 'Ye Old Shoppe'. Ye is really pronounced 'The'.

The other is the 'long S' that looks a little like a lower case 'F', it was commonly used in Shakespeare's time.
