I think the Nephilim were the children that came of the union of the Sons of God and the Daughters of Man, and for some reason they were giants or something. Something went haywire genetically. And the whole 12 fingers, 12 toes bit too.
I've heard some people suggest that the humans mated with angels, but I don't get that from this particular passage. I'm no Bible scholar, so I don't know.
