Pre-exposure to mRNA-LNP inhibits adaptive immune responses and alters innate immune fitness in an inheritable fashion

 

ABSTRACT

Hundreds of millions of SARS-CoV-2 mRNA-LNP vaccine doses have already been administered to humans. However,we lack a comprehensive understanding of the immune effects of this platform.The mRNA-LNP-based SARS-CoV-2 vaccine is highly inflammatory, and its synthetic ionizable lipid component responsible for the induction of inflammation has a long in vivo half-life. Since chronic inflammation can lead to immune exhaustion and non-responsiveness, we sought to determine the effects of pre-exposure to the mRNA-LNP on adaptive immune responses and innate immune fitness. We found that pre-exposure to mRNA-LNPs or LNP alone led to long-term inhibition of the adaptive immune responses, which could be overcome using standard adjuvants. On the other hand, we report that after pre-exposure to mRNA-LNPs, the resistance of mice to heterologous infections with influenza virus increased while Candida albicans decreased. The diminished resistance to Candida albicans correlated with a general decrease in blood neutrophil percentages. Interestingly, mice pre-exposed to the mRNA-LNP platform can pass down the acquired immune traits to their offspring, providing better protection against influenza. In summary, the mRNA-LNP vaccine platform induces long-term unexpected immunological changes affecting both adaptive immune responses and heterologous protection against infections. Thus, our studies highlight the need for more research to determine this platform’s true impact on human health.

 

Authors Summary We bring experimental evidence that pre-exposure to mRNA-LNPs or its LNP component affects innate and adaptive immune responses. Pre-exposure to mRNA-LNPs led to long-term inhibition of the adaptive immune responses, which the use of adjuvants could overcome. More studies are needed to understand the mechanisms responsible for these effects and determine this platform’s impact on human health.

I hope this isn’t too controversial. It’s certainly frightening, but I believe we are still on the right side of disaster & if enough of us become aware of what is happening here & everywhere in the democratic world, we can recover the situation. We really don’t have long. I believe it’s likely things will change irretrievably over this coming winter. Hence this urgent and unusual request.

 

Everything that’s happened and is happening becomes much simpler and it all makes sense, only when you force yourself to think the impossible.

 

If you experimentally adopt the position that OUR GOVERNMENT IS ACTIVELY WORKING TO HARM US, TO DISMANTLE MODERN SOCIETY & ENSLAVE ALL PEOPLE IN A DIGITALLY CONTROLLED TOTALITARIAN WORLD, it all fits. Nothing is surplus.

 

Even if your immediate response is that this is absurd, please try it for a day or so.

 

I ask you further to adopt the experimental position that the media, controlled by just six global corporations, all allied to a single global organization you’ve all heard of, is relentlessly lying to you and has been doing so for over 2.5 years. Same for the internet, controlled by fewer global corporations, also all allied to that same global organization.

 

Because I am certain it’s true. I am certain because this all started with a scientific fraud relating to a virus, augmented it with a relentless campaign of fear, imposed measures known to be useless, which wrecked the economy and smashed civil society, then coerced most to accept useless, unnecessary, ineffective and deliberately dangerous injections. Obviously, this is an odious crime. Nothing like it has ever happened.

The Godfather

In a pivotal scene in the novel and film, Fredo is with his father when assassins working for drug kingpin Virgil Sollozzo (Al Lettieri) gun down Don Corleone in the street. Fredo, terrified, drops his gun, failing to return fire. He sits on the curb next to his severely wounded father, weeping. In the novel, Fredo is sickened after witnessing his father being shot, going into shock. To aid Fredo's recovery and protect him from possible reprisals, Sonny sends his younger brother to Las Vegas under the protection of Don Anthony Molinari of San Francisco.

 

The Godfather Part II

While in Havana negotiating with Roth, Michael discovers that Fredo is the family traitor behind the assassination attempt on him. After telling Michael that he has never met Ola, Fredo later carelessly tells Geary that he had been to a nightclub with Ola. Michael overhears the conversation and realizes that Fredo is the traitor within the family. He confronts Fredo, delivering the kiss of death. Amid the chaos of American-backed dictator Fulgencio Batista fleeing Fidel Castro's rebel army, Michael pleads with Fredo to leave the country with him. Frightened, Fredo runs away into the crowd. Michael's men eventually locate Fredo and convince him to return home.

 

Michael is indicted by a Senate subcommittee investigating organized crime. Michael's former caporegime, Frank Pentangeli (Michael V. Gazzo), is scheduled to testify against Michael at the hearing. A few days before the hearing, Michael asks Fredo what he knows regarding Roth's plans. Fredo claims that he did not know they would make an attempt on Michael's life, and that if he helped Roth, "there was something in it for me, on my own". He tells Michael that he resents being passed over to succeed their father; he believes that, as the older brother, he should have taken over the family business after Vito's death. When pressed by Michael, Fredo reveals that the Senate commission's lawyer is on Roth's payroll. Michael disowns Fredo, and privately instructs his personal assassin Al Neri (Richard Bright) that nothing is to happen to Fredo while their mother is alive; the implication being that Fredo will be killed after her death. At their mother's funeral, and at their sister Connie's urging, Michael forgives Fredo based on this conversation with Connie. This agreement was soon broken when Connie let Michael's ex-wife see their children in secret and behind his back. Soon after, while Fredo and Neri are fishing on Lake Tahoe, Neri executes Fredo as he is praying the Hail Mary, as Michael watches from his house.

 

The Godfather Part III

Fredo appears only once in the third film, in a flashback depicting his death through archive footage. He is also mentioned many times throughout the film; the dialogue makes it clear that Michael is tormented with guilt over ordering his brother's death, and that it has alienated him from his ex-wife, Kay (Diane Keaton), and his son, Anthony (Franc D'Ambrosio), both of whom know that Michael ordered Fredo's death. The official explanation of Fredo's death, as related by Connie, is that he drowned, although it is left ambiguous whether Connie actually believes this. Michael himself cries out Fredo's name while having a diabetic stroke. Later in the film, he breaks down in tears while confessing having ordered Fredo's death to Cardinal Lamberto (Raf Vallone), who later becomes Pope John Paul I.

China Locks Down Megacity Chengdu as Covid Zero Intensifies

City of 21 million is biggest to be locked down since Shanghai

Electronics, automaker supply chains at risk of disruption

 

The Chinese metropolis of Chengdu locked down its 21 million residents to contain a Covid-19 outbreak, a seismic move in the country’s vast Western region that has largely been untouched by the virus.

 

The capital of Sichuan province, Chengdu is the biggest city to shut down since Shanghai’s bruising two-month lockdown earlier this year. The move which will upend the lives of millions of people and businesses, with repercussions for China’s economy and beyond shows the country’s commitment to the Covid Zero approach espoused by President Xi Jinping, despite the disruption it’s causing.

The lockdown of Chengdu Thursday came after 157 new cases were reported. The city, which accounts for about 1.7% of China’s gross domestic product, is home to numerous technology companies and automakers. Foxconn Technology Group, the world’s largest assembler of Apple Inc.’s iPhones, is moving to a closed-loop manufacturing system for the iPads it makes there, according to a person familiar with the decision, while Volvo Car AB suspended work at its factory.

 

Chengdu is also a popular tourist destination, famed for its giant panda sanctuary.

 

Chengdu Lockdown

The entire city will undergo four days of mass testing starting Thursday evening. The extent of the economic dislocation depends on the duration of the lockdown, which city officials haven’t yet disclosed. Some places in China have imposed snap closures lasting a few days or even just hours, while others extended isolation rules until their outbreaks were fully under control.

 

While consumer activities like shopping and dining out will be impacted immediately, the fallout could be severe for the region’s manufacturing sector if supply chains are severed for a protracted period, depleting factory inventories and hitting production. The lockdown’s impact on sentiment will likely amplify the damage, dealing another major blow to China’s growth, David Qu, an economist at Bloomberg Economics, wrote in a report.

 

Shenzhen’s Contribution

Mounting restrictions in the southern technology hub Shenzhen could compound the pain for China, as the risk of a lockdown of the more economically important city and its 12.6 million people grows along with its Covid cluster. Cases nearly doubled overnight to 62, even as several districts closed venues and ordered residents to stay home.

 

The lockdown component of the Covid Zero approach is being embraced nationwide thanks to its success in snuffing out cases, which is still China’s goal more than two years into the pandemic. There’s additional pressure to tame flareups in the run up to the Communist Party congress later this year, when Xi is expected to secure a precedent-breaking third term as leader.

Global stocks hit a six-week low on news of Chengdu’s lockdown and hawkish commentary from central banks. A gauge of Chinese stocks listed in Hong Kong extended declines to end Thursday’s session with a 1.8% loss, its biggest in three weeks. China’s benchmark CSI 300 Index slid 0.9% to close at a three-month low. Currencies in emerging markets also fell, with the South Korean won dropping more than 1% against the dollar as the news spurred more risk aversion.

 

Panic Buying

Residents, meanwhile, are bracing for the worst.

 

Shanghai’s two-month lockdown of its 25 million residents is front-of-mind for many people. It too started with a four-day testing drive. While half of the city that accounts for 3.8% of the country’s gross domestic product initially locked down in late March, it didn’t reopen as expected when the other half shut.

 

Chloe Liu, a 25-year-old dancer, said signs of the food shortages that besieged Shanghai are already appearing in Chengdu. Grocery apps are showing insufficient delivery capacity across the city, with some entirely out of stock.

“I feel the sort of craziness for the first time at a grocery market when I just went downstairs trying to stock up some food,” she said. “Literally everyone is panic buying.”

 

Despite the Covid Zero approach, the virus continues to flare within China because of its increasing contagiousness. The aggressive measures typically work, but need to be deployed more often as infections pop up in different parts of the country.

Total infections in Chengdu in the current outbreak have exceeded 900, according to a tally of daily numbers released by the local health commission.

 

Residents must stay in their homes, except for exceptional situations, such as a medical emergency, the city’s government said. Families can send one person out, once a day, for groceries. People can leave Chengdu if they present a negative Covid test result taken within the previous 24 hours.

 

Weather Impact

Due to extremely hot weather in the city, frequent contact among people in indoor entertainment venues including water parks led to increased transmission of the virus, Yang Xiaoguang, head of Chengdu’s health commission, told reporters on Tuesday.

 

Chengdu postponed the start of the school year on Wednesday, as did multiple cities across China, citing the need to prevent virus transmission.
