Artificially Expanded Genetic Information System (AEGIS) is a synthetic DNA analog experiment that uses some unnatural base pairs from the laboratories of the Foundation for Applied Molecular Evolution in Gainesville, Florida. AEGIS is a NASA-funded project to try to understand how extraterrestrial life may have developed.

The system uses twelve different nucleobases in its genetic code. These include the four canonical nucleobases found in DNA (adenine, cytosine, guanine and thymine) plus eight synthetic nucleobases). AEGIS includes S:B, Z:P, V:J and K:X base pairs.

Hachimoji DNA ("eight letters") is a synthetic nucleic acid analog that uses four synthetic nucleotides in addition to the four present in the natural nucleic acids, DNA and RNA. This leads to four allowed base pairs: two unnatural base pairs formed by the synthetic nucleobases in addition to the two normal pairs. Hachimoji bases have been demonstrated in both DNA and RNA analogs, using deoxyribose and ribose respectively as the backbone sugar.

Benefits of such a nucleic acid system may include an enhanced ability to store data, as well as insights into what may be possible in the search for extraterrestrial life.

The hachimoji DNA system produced one type of catalytic RNA (ribozyme or aptamer) in vitro.

One of the most promising developments lies in a beaker of water inside a Florida laboratory. It's an experiment called AEGIS — an acronym for Artificially Expanded Genetic Information System. Its creator, Steve Benner, says it is the first synthetic genetic system capable of Darwinian evolution.

AEGIS is not self-sustaining, at least not yet, and with 12 DNA building blocks — as opposed to the usual four — there's little chance it will be confused with natural life. Still, Benner is encouraged by the results.

"It's evolving. It's doing what we designed it to do," said Benner, a biochemist with the Gainesville, Fla.-based Foundation for Applied Molecular Evolution.

OligArch: A software tool to allow artificially expanded genetic information systems (AEGIS) to guide the autonomous self-assembly of long DNA constructs from multiple DNA single strands

Synthetic biologists wishing to self-assemble large DNA (L-DNA) constructs from small DNA fragments made by automated synthesis need fragments that hybridize predictably. Such predictability is difficult to obtain with nucleotides built from just the four standard nucleotides. Natural DNA's peculiar combination of strong and weak G:C and A:T pairs, the context-dependence of the strengths of those pairs, unimolecular strand folding that competes with desired interstrand hybridization, and non-Watson–Crick interactions available to standard DNA, all contribute to this unpredictability. In principle, adding extra nucleotides to the genetic alphabet can improve the predictability and reliability of autonomous DNA self-assembly, simply by increasing the information density of oligonucleotide sequences. These extra nucleotides are now available as parts of artificially expanded genetic information systems (AEGIS), and tools are now available to generate entirely standard DNA from AEGIS DNA during PCR amplification. Here, we describe the OligArch (for "oligonucleotide architecting") software, an application that permits synthetic biologists to engineer optimally self-assembling DNA constructs from both six- and eight-letter AEGIS alphabets. This software has been used to design oligonucleotides that self-assemble to form complete genes from 20 or more single-stranded synthetic oligonucleotides. OligArch is therefore a key element of a scalable and integrated infrastructure for the rapid and designed engineering of biology.

Because of chemical behaviors of real DNA molecules, the nanotechnology envisioned by the Defense Department in this Solicitation will be possible only if design automation software creates nanostructures that exploit artificially expanded genetic information systems (AEGIS). AEGIS DNA molecules have more than the four nucleotides (GACT) found in natural DNA. In Phase 1, we delivered a demonstration project showing that adding nucleotides to standard DNA supports three-dimensional self-assembling nanoarchitectures at the frontier of the field. We also improved several of the AEGIS components. This progress will allow Phase 2 to deliver kits for specialists, non-specialists, and students that combine AEGIS DNA with non-DNA linkers to give a nano AEGIS"Lego"kit. We will also deliver a nanostructure as a signaling element to detect DNA from insect-borne pathogens that cause rickettsial diseases (e.g. typhus). This project overlaps Firebird's business, which supports human diagnostics based on reagent innovations like AEGIS. Adding nanotechnology to its existing business makes commercial sense, and will help"dual uses"emerge from this project.

WASHINGTON (AP) — Vice President Joe Biden is praising Jewish leaders for helping change American attitudes about gay marriage and other issues.

Biden says culture and arts change people’s attitudes. He cites social media and the old NBC TV series “Will and Grace” as examples of what helped changed attitudes on gay marriage.

Biden says, quote, “Think — behind of all that, I bet you 85 percent of those changes, whether it’s in Hollywood or social media, are a consequence of Jewish leaders in the industry.”

Biden says the influence is immense and that those changes have been for the good.

Biden was speaking Tuesday night at a Jewish American Heritage Month reception hosted by the Democratic National Committee. He says Jewish values are an essential part of who Americans are.

Miller confirmed with the Daily Caller that the staffer’s name is Aidan Maese-Czeropski. According to Maese-Czeropski’s LinkedIn profile, he serves as a legislative aide for Democratic Maryland Sen. Ben Cardin. Sources tipped off the Caller that the staffer, who confronted Miller during an interview with NBC News’ Ali Vitali and said “Free Palestine!,” served as an aide for Cardin.

The Caller contacted Cardin’s office about Maese-Czeropski confronting Miller, to which they replied, “Are you sure you have the correct person? Having seen press reports, I understood this was a House staffer. Also, the person you mention is much more junior than an LD, so perhaps there was a mix up in identification, as well? Additional information would be of help.”

The Caller then told Cardin’s office that Miller confirmed his identity, to which they did not respond.
