Methylene blue (MB) has successfully been used to treat malaria (Rengelshausen et al., 2004, Zoungrana et al., 2008, Coulibaly et al., 2009, Krafts et al., 2012), ifosfamide neurotoxicity (Raj et al., 2004, Patel, 2006, Richards et al., 2011) and methemoglobinemia (Dotsch et al., 2000, Zenk, 2001, Matisoff and Panni, 2006). Moreover, recent studies have suggested that MB has an important neuroprotective effect on Parkinson's disease and ischemic stroke through its ability to act as an optional mitochondrial electron carrier (Wen et al., 2011, Congdon et al., 2012). Studies have also shown that MB activates AMP-activated protein kinase (AMPK) and silent mating-type information regulation 2 homolog 1 (SIRT1) when used to treat steatosis and steatohepatitis (Shin et al., 2014). Nevertheless, neither the ability of MB treatment to alleviate the symptoms of EAE nor the mechanism underlying its effects has been explored.
