If you cannot follow an order or a policy, etc you resign and you are not waiting for it to be accepted, or not accepted, by your boss. In politics this type of resignation is to bring attention to a policy or order with which the resignor disagrees.
There is also the conditional resignation. It can be used to bolster power/leverage. In this scenario the resignor is gambling that the boss will back his play and empower him over his rivals.
Then there is the garden variety resignation. Every appointee writes a resignation letter as a precondition to accepting the appointment. If the boss is dissatisfied, in any way, with his performance he will pull out the pre signed resignation and accept the same.
