No on cloture doesn't mean no on the actual vote. Cloture is to limit the debate; she may feel more debate is needed. But tomorrow faced with up or down, she could well vote yes. Conversely Manchin voting yes on cloture signals he doesn't think any more debate is needed (which would make sense given his other comments); doesn't mean we will vote yes to confirm (although I suspect he will).
