In this review, we describe the physiological and pathophysiological substrates of furin and discuss how dysregulation of a simple proteolytic cleavage event may promote infectious diseases and cancer.

Targeted RNA recombination has proven to be a powerful tool for the genetic engineering of the coronavirus genome

Key to the two-step method is the efficient selection of recombinant viruses based on host cell switching.

The first step consists of the preparation -using this selection principle- of an interspecies chimeric coronavirus. In this virus the ectodomain of the spike glycoprotein is replaced by that of a coronavirus with a different species tropism.

In the second step this chimeric virus is used as the recipient for recombination with synthetic donor RNA carrying the original spike gene. Recombinant viruses are then isolated on the basis of their regained natural (e.g., murine or feline or bats) cell tropism.

Additional mutations created in the donor RNA can be co-incorporated into the recombinant virus in order to generate mutant viruses.

The fact that COVID-19 has cleavage sites for furin enzymes means that this virus can be extremely pathogenic, with the capacity to replicate in MULTIPLE tissues and organs due to how furins are utilized and distributed in the human body [7].

 

In addition, bioweapons such as Anthrax require furin to initiate toxicity. Furin has been identified as the cellular protease that cleaves the protective antigen protein of anthrax toxins that unleashes toxic activity [10].

 

In a similar way, envelope proteins of viruses including HIV, influenza, dengue fever, ebola, and marburg virus, MUST be cleaved by furin or furin-like proteases to become fully active and functional. COVID-19, a novel coronavirus that originated from Wuhan, China, has now been identified to possess furin-cleavage sites in the S protein.

 

Thus COVID-19 and a host of important substrates found in the human body, including cytokines, hormones, growth factors and receptor all need to be cleaved by furin enzymes for activation. It is now becoming much clearer why furin-cleavage sites in viruses can increase their virulence and pathogenicity.

Would be interesting to see if there is an anomaly in the number of VAP coded cases.

You would think that those would have risen recently also since there was an increase of patients placed on a ventilator.
