Ackchyually; it should be a semicolon! The term "bitch" is essentially a stand alone sentence but is tied to the beginning part so; rather than a comma, which is a soft pause, a semicolon provides a hard pause but not as hard as a full stop thereby remaining attached to the complete sentence while still conveying the emphasis required of the phrase.

It would be:

"I don't think so; bitch."

When a semicolon is used, the word following is never capitalised unless it is a name. The semicolon merely gives an increased pause, greater than a comma but, shorter than a full stop. Period for the American grammar students.
