Base editing: Revolutionary therapy clears girl's incurable cancer

A teenage girl's incurable cancer has been cleared from her body in the first use of a revolutionary new type of medicine.

All other treatments for Alyssa's leukaemia had failed.

So doctors at Great Ormond Street Hospital used "base editing" to perform a feat of biological engineering to build her a new living drug.

Six months later the cancer is undetectable, but Alyssa is still being monitored in case it comes back.

Alyssa, who is 13 and from Leicester, was diagnosed with T-cell acute lymphoblastic leukaemia in May last year.

T-cells are supposed to be the body's guardians - seeking out and destroying threats - but for Alyssa they had become the danger and were growing out of control.

Her cancer was aggressive. Chemotherapy, and then a bone-marrow transplant, were unable to rid it from her body.

Without the experimental medicine, the only option left would have been merely to make Alyssa as comfortable as possible.

"Eventually I would have passed away," said Alyssa. Her mum, Kiona, said this time last year she had been dreading Christmas, "thinking this is our last with her". And then she "just cried" through her daughter's 13th birthday in January.

What happened next would have been unthinkable just a few years ago and has been made possible by incredible advances in genetics.

The team at Great Ormond Street used a technology called base editing, which was invented only six years ago.

Bases are the language of life. The four types of base - adenine (A), cytosine (C), guanine (G) and thymine (T) - are the building blocks of our genetic code. Just as letters in the alphabet spell out words that carry meaning, the billions of bases in our DNA spell out the instruction manual for our body.

Base editing allows scientists to zoom to a precise part of the genetic code and then alter the molecular structure of just one base, converting it into another and changing the genetic instructions.

The large team of doctors and scientists used this tool to engineer a new type of T-cell that was capable of hunting down and killing Alyssa's cancerous T-cells.

They started with healthy T-cells that came from a donor and set about modifying them.

The first base edit disabled the T-cells targeting mechanism so they would not assault Alyssa's body

The second removed a chemical marking, called CD7, which is on all T-cells

The third edit was an invisibility cloak that prevented the cells being killed by a chemotherapy drug

The final stage of genetic modification instructed the T-cells to go hunting for anything with the CD7 marking on it so that it would destroy every T-cell in her body - including the cancerous ones. That's why this marking has to be removed from the therapy - otherwise it would just destroy itself.

If the therapy works, Alyssa's immune system - including T-cells - will be rebuilt with the second bone-marrow transplant.

Former Twitter exec Yoel Roth argued for minors’ access to adult internet services in PhD thesis

"…it's worth considering how, if at all, the current generation of popular sites of gay networked sociability might fit into an overall queer social landscape that increasingly includes individuals under the age of 18."

On Saturday Twitter CEO Elon Musk called out former head of Twitter Trust and Safety Yoel Roth for "arguing in favor of children being able to access adult Internet services in his PhD thesis."

Musk tweeted an excerpt from Roth's thesis, portions of which read, "…it's worth considering how, if at all, the current generation of popular sites of gay networked sociability might fit into an overall queer social landscape that increasingly includes individuals under the age of 18."

"The fact that people under 18 are on these services already indicates that we can't readily dismiss these platforms out of hand as loci for queer youth culture," Roth said. "Rather than merely trying to absolve themselves of legal responsibility or, worse, trying to drive out teenagers entirely, service providers should instead focus on crafting safety strategies that can accommodate a wide variety of use cases."

Musk was responding to a tweet from Eliza Bleu that linked to a Roth tweet from 2010 that read, "Can high school students ever meaningfully consent to sex with their teachers?" Roth also linked to a Salon article titled "Student-teacher sex: When is it OK?" 

The Salon piece documents the story of a High School choir teacher, Matthew Hirschfelder, who had sex with a student. According to Salon, "Things get messier when looking at cases like Hirschfelder's. If we draw the line at high school graduation, as opposed to at a student's 18th birthday, it means that a teacher having sex with a 20-year-old high school student could be outlawed."

Fox News producer Gregg Re reached out to the Annenberg School for Communication at Penn State University, asking for comment as to Roth's controversial thesis.

"You awarded a deviant named @yoyoel a PHD," Re said along with his inquiry.

The Post Millennial reported about Twitter's child pornography problem in 2020 and how prolific sharing sexual images of minors had become on the site. When Elon Musk bought Twitter, he said it was "Priority #1" to eliminate child porn on the platform and worked to erase the hashtags users were exploiting to help spread the images.

Roth quit soon after.

On November 20, Roth published an op-ed with the New York Times and said he fought "dangerous misinformation" while at the social media giant, and said, "yes, ban[ning] President Donald Trump" was part of his efforts. He also said Musk's goal of "unfettered speech" was not right for the platform.

Twitter users called Roth out then for how he allowed child pornography to flourish at the outlet. One user wrote, "Why was child pornography permitted on Twitter for years under your watch, but stamped out within 48 hrs after you left?"

The multiple rounds of the "Twitter Files" confirmed that Roth was complicit in censorship of conservative voices on the site. In early December, Roth defended banning the Babylon Bee and Libs of TikTok on Twitter and said the users make "people unsafe in the world."

Chile's Lascar volcano erupts for first time in 30 years

A volcano in the Andes in Chile's north rumbled to life on Saturday, triggering minor earth tremors and sending a plume of smoke and ash thousands of metres into the sky.

Chile's National Geology and Mining Service reported that the Lascar volcano stirred at 12.36pm.

The volcano sent "an eruptive column" of volcanic ash and hot gases 6,000 metres above its crater, the service said.

Authorities raised an alert level to yellow, indicating elevated volcanic activity, and established a no-entry perimeter 5km around the crater.

They also alerted air traffic to the drifting plume.

Authorities stayed in close contact with officials in Talabre, a town 30km from the volcano, in case evacuations were required. But no property damage was reported.

Lascar, with an elevation of 5,592m above sea level, is 70km from San Pedro de Atacama, a popular tourist centre that draws visitors for trekking, amateur astronomy and visits to the Atacama Desert, the driest place on Earth.

Lascar erupted in 1993 but also displayed lesser volcanic activity in 2006 and 2015.

Farther to the south, yellow alerts remain in effect for regions around the Nevados de Chillan volcanic complex and the Villarrica volcano.

“Many of those misbehaving today are too young to be aware of the destruction and loss of lives that occurred during the Nigerian Civil War. Those of us in the fields for 30 months, who went through the war, will treat them in the language they understand,” Buhari wrote in the now-deleted tweet, referring to the brutal two-year Nigeria-Biafra war, which killed an estimated one to three million people, mostly from the Igbo tribe in the eastern part of the country between 1967-1970.

The tweet was deleted Wednesday after many Nigerians flagged it to Twitter and the platform said it had violated its policy on abusive behavior.

Information Minister Lai Mohammed criticized Twitter’s action and accused the social media giant of “double standards.”

Mohammed also questioned Twitter’s motives in Nigeria, saying, “the mission of Twitter in Nigeria is very very suspect…” at a news conference on Wednesday after Buhari’s tweet was deleted.
