The discussion about covid response is often clouded by a conflation of 2020 and 2021.

 

In 2020, the unceasing attack against Trump, including from countless Republicans, was that he wouldn’t listen to experts, wouldn’t impose fed mandates, wouldn’t stop pushing alternative therapies, wouldn’t back the “science,” and so on, ad infinitum. (Polling at this time sadly showed all kinds of support for profoundly restrictive federal requirements that Trump refused to impose at the height of the hysteria).

 

PPP loans were a response to international, state, city, local and corporate covid shutdowns — not federal ones.

 

Indeed, Biden “campaigned” on the idea that he would heed the very “experts” Trump had contradicted and shunned.

 

In 2021, Biden put these “experts” in charge. Soon after, Republican state officials and federal lawmakers mounted a resistance, following a growing public revolt against Democrats’ ruinous mandates, along with the piercing of the oppressive covid censorship regime.

(Headline) Optogenetics: Controlling the Brain with Light

 

Optogenetics is the combination of genetics and optics to control well-defined events within specific cells of living tissue. It includes the discovery and insertion into cells of genes that confer light responsiveness; it also includes the associated technologies for delivering light deep into organisms as complex as freely moving mammals, for targeting light-sensitivity to cells of interest, and for assessing specific readouts, or effects, of this optical control.

 

What excites neuroscientists about optogenetics is control over defined events within defined cell types at defined times—a level of precision that is most likely crucial to biological understanding even beyond neuroscience. The significance of any event in a cell has full meaning only in the context of the other events occurring around it in the rest of the tissue, the whole organism or even the larger environment. Even a shift of a few milliseconds in the timing of a neuron's firing, for example, can sometimes completely reverse the effect of its signal on the rest of the nervous system. And millisecond-scale timing precision within behaving mammals has been essential for key insights into both normal brain function and into clinical problems such as parkinsonism.

Fluorescent proteins

 

The multicolored fluorescent proteins developed in Tsien's lab are used by scientists to track where and when certain genes are expressed in cells or in whole organisms. Typically, the gene coding for a protein of interest is fused with the gene for a fluorescent protein, which causes the protein of interest to glow inside the cell when the cell is irradiated with a suitable wavelength of light and allows microscopists to track its location in real time. This is such a popular technique that it has added a new dimension to the fields of molecular biology, cell biology, and biochemistry.[9]

 

Since the discovery of the wild type GFP, numerous different mutants of GFP have been engineered and tested.[23] The first significant leap forward was a single point mutation (S65T) reported by Tsien in 1995 in Nature.[24] This mutation dramatically improved the fluorescent (both intensity and photostability) and spectral characteristics of GFP. A shift of the major excitation peak to 488 nm with the emission peak staying at 509 nm thus can be clearly observed, which matched very well the spectral characteristics of commonly available FITC facilities.

Fluorophores for the Blue (488 nm) Laser

 

Overview

The blue laser is versatile due to its ability to excite both FITC (and similar fluorophores), PE, and PE tandems. PE is also excitable by the 561 nm laser and so PE based fluorophores appear on both lists. However, as PE is maximally excited at 546 nm, where possible it is advisable to use the 561 nm laser to get the maximal signal and to reduce spillover and therefore compensation. This also frees up space on the blue laser for additional fluorophores.

With similar fluorescence properties to FITC, Alexa Fluor 488 is not compatible with FITC or GFP. However, as it is brighter and more photostable, it is a great alternative, especially for intracellular staining and in fluorescence microscopy.

Investigation Into Biden, Classified Docs Is Reportedly Far Wider Than Previously Believed

Federal prosecutors and FBI agents have significantly widened the investigation into President Joe Biden’s handling of classified documents, according to ABC News.

 

Prosecutors and agents from Special Counsel Robert Hur’s office haveinterviewed as many as 100 witnesses, including Secretary of State Antony Blinken, and have examined internal White House processes during the past nine months, according to the outlet. Some interviews were conducted as late as mid-September, and some interviewees have even been asked to return for a follow-up interview.

 

Authorities have reportedly gathered information regarding Biden’s alleged carelessness in the handling of classified material during his time as vice president, ABC News reported. The witnesses reportedly said the removal of the documents appeared to be a mistake rather than criminal activity.

 

In the course of the investigation, Hur’s office has been accessing and investigating documents dating back to the early days of former President Barack Obama’s administration and Biden’s tenure in the Senate, according to ABC News.

 

Witnesses have reportedly been questioned about Biden and his aides’ internal procedures for handling classified material, especially where Biden stored documents, notecards and briefing books, ABC News reported.

 

They have also accessed email chains dating back to 2010 and have asked witnesses for context and have reportedly asked witnesses about the use of safes and cabinets, per the outlet.

 

Biden’s lawyers found ten classified documents at the University of Pennsylvania’s Biden Penn Center on Nov. 2, leading to an FBI search. More documents were found at his private home in Delaware on Dec. 20, Jan. 11, Jan. 12 and Jan. 20.

 

Biden’s attorneys did not announce the discovery of the first tranche of documents until January, though they initially discovered them just ten days before the 2022 midterm elections.

 

Attorney General Merrick Garland appointed Hur as special counsel on Jan. 12 to investigate whether the president mishandled the classified documents.

A former adviser to former President Barack Obama blasted President Joe Biden for joining striking members of the United Auto Workers (UAW) Tuesday.

 

Biden attended a picket line in Michigan Tuesday, the day before former President Donald Trump was slated to give a major speech in that state instead of attending a Republican primary debate. The UAW walked off the job at three auto manufacturing plants in Ohio, Michigan and Missouri at midnight Sept. 15 after failing to reach an agreement with Ford, General Motors and Stellantis (formerly Chrysler).

 

“For him to be going on a picket line is outrageous,” Steve Rattner, who was the “auto czar” during the Obama administration, said in an interview, according to NBC News. “There’s no precedent for it. The tradition of the president is to stay neutral in these things. I get the politics. The progressives all said, ‘We don’t want a mediator; we want an advocate.’ Andhe bowed to the progressives, and now he’s going out there to put his thumb on the scale. And it’s wrong.”

“You deserve what you’ve earned and you’ve earned a hell of a lot more than you gettin’ paid now,” Biden said during his visit to the picket line.

 

The union sought a four-day work week and a 36% salary increase over five years, accordingto Bloomberg. Biden endorsed the union’s demand for the pay increase. The UAW has also criticized President Joe Biden’s efforts to promote electric vehicles, Politico reported.

Over the course of the first of the Mad Max movies, the young officer's wife and child are brutally murdered by one of these road gangs, sending him on a mission of vengeance; he hunts down each member in his trademark interceptor and kills them one by one. In the Mad Max:Fury Road prequelcomics, however, this is changed,and the nuclear war takes place between the first and second films. Regardless of the timeline, the effect is the same: society has continued to break down significantly in just a few short years since the end of Mad Max, especially in the Australian wasteland where Max now lives and travels around in his interceptor searching for gasoline.

 

There are forms of civilization left, including a peaceful tribe of people that Max ends up joining. I introduced channelrhodopsin-2 into mammalian neurons in culture by the well-established techniques of transfection—that is, by splicing the gene for ChR2 and a specific kind of on switch, or promoter, into a vector (like a benign virus) that ferried the added genetic material into the cells. Promoters can ensure that only selected kinds of neurons (such as those able to secrete the neurotransmitter glutamate) will express, or make, the encoded proteins.

 

Against all odds, the experiments worked shockingly well. Using nothing more than safe pulses of visible light, we attained reliable, millisecond-precision control over the cells' patterns of firing of action potentials—the voltage blips, or impulses, that enable one neuron to convey information to another. In August 2005 my team published the first report that by introducing a microbial opsin gene, we could make neurons precisely responsive to light. Channelrhodopsins (and, eventually as we found, the bacteriorhodopsin from 1971 and the halorhodopsins, too) all proved able to turn neurons on or off, efficiently and safely in response to light. They worked in part because mammalian tissues contain naturally robust quantities of all-trans retinal—the one chemical cofactor essential for photons to activate microbial opsins—so nothing beyond an opsin gene needs to be added to targeted neurons. The number of optogenetic tools, along with the diversity of their capabilities, has since expanded rapidly because of a remarkable convergence of ecology and engineering. Investigators are adding new opsins to their tool kits by scouring the natural world for novel ones; they are also applying molecular engineering to tweak the known opsins to make them even more useful for diverse experiments in a wider range of organisms.

 

In 2008, for instance, our genome searches led by Feng Zhang on a different algal species, Volvox carteri, revealed a third channelrhodopsin (VChR1), which responds to yellow light instead of blue as we showed together with Peter Hegemann. Using VChR1 and the other channelrhodopsins together, we can simultaneously control mixed populations of cells, with yellow light exerting one type of control over some of them and blue light sending a different command to others. And we now have found that the most potent channelrhodopsin of all is actually a hybrid of VChR1 and ChR1 (with no contribution from ChR2 at all). Our other modified opsins (created with Ofer Yizhar, Lief Fenno, Lisa Gunaydin and Hegemann and his students) now include "fast" and "slow" channelrhodopsin mutants that offer exquisite control over the timing and duration of action potentials: the former can drive action potentials more than 200 times per second, whereas the latter can push cells into or out of stable excitable states with single pulses of light. Our newest opsins can also now respond to deep red light that borders on the infrared, which stays more sharply focused, penetrates tissues more easily and is very well tolerated by subjects. Many groups are now also pushing opsin engineering forward, including those of Hiromu Yawo in Japan, Ernst Bamberg in Frankfurt and Roger Tsien in San Diego.

The person then goes and sacrifices some of what he has. Ukrainians sacrificed their constitution in 2004. A pro-Western presidential candidate lost the elections but they made him president anyway.

Things have only been getting worse. But the person thinks: "I need to gamble more, that way I increase my chances of winning. I don't have much but I'll spend some more."

Bit by bit, Ukrainians have given away their remaining sovereignty, the existing legal order, the economy. They broke the economic ties with Russia and became the poorest country in Europe.

At some point, the frustration builds up: the person spent all his money gambling but he hasn't won that big prize.

The frustration is so powerful that he cannot stop. He thinks: "I lost so much already and if I stop now, it means I wasted it all".

Remember: this person from the very beginning did not want to work hard to improve his life and accept the fact that, perhaps, he won't be a millionaire, but he could have a decent, stable life. Now, after he lost so much, he's even less likely to do it, because he'd have to work even harder.

That's when the addiction devours the person completely. Don't ukies all have somewhat of an insane look? They take everything to the "casino": the memory of their grandparents who fought against Nazism. They start paying a mandatory "war tax" in 2015 to fight against the Donbass. They are killed and injured by thousands not only at the battlefield, but also on the Ukrainian territory, by Nazis and conscriptors. This doesn't stop the addicts, they don't even express much concern about it.

Even more frantically than before, they believe that by sacrificing it all, they will win the big prize. They will become rich and happy.

Along the way, the "casino" lets them win little pennies, incomparable to what they lost, but in the sick addict's mind it reinforces his faith that some day, one day, he will be lucky and get himself his dream life.

All the developing countries should look at this and learn. These psychological mechanisms are most likely deliberately applied by the color revolution architects.

With sound as the culprit, at the right frequency, amplitude and duration, health may be at risk. Go to a heavy-metal concert for an hour or so, it is hoped without ear damage. However, there might be nausea as a side effect. In that situation, sound has made the body vibrate and react.

 

The span of sound that a person hears is termed frequency range; the unit of measurement, Hertz (hz). Although there is considerable variation between individuals, the hearing range is commonly accepted to be 20 to 20,000 hz. To place that in perspective, the low frequency of a Tuba is 29 hz and of a Bass 27 hz. Below 20 hz, it’s called “Infrasound.” Those sounds are imperceptible to the human ear, but the body hears it, although, one may not be aware of the bombardment.

 

These sounds might occur from the whir of motors, water pumps, construction site noise, equipment room near your residence, or close-by traffic. Daria Vaisman, a research editor at the New York Press told of an incident with Walt Disney and his team of cartoonists. They slowed down a 60-cycle tone in a short cartoon to 12 Hz; they became sick for days afterward. A good example of extreme low frequencies that might be encountered is the church pipe organ. It can cause sensations of sorrow, coldness, anxiety and even shivers down the spine.

Attorneys for Man Charged With Killing 2 Teenage Indiana Girls Argue They Died in Ritual Sacrifice

DELPHI, Ind.—Attorneys for a northern Indiana man charged with killing two teenage girls contend in court documents filed Monday that their client had nothing to do with the crimes and that the girls actually died as part of a ritual sacrifice.Attorneys Andrew Baldwin and Brad Rozzi, who represent Richard Allen of Delphi, said in a 136-page document that 13-year-old Abigail Williams and 14-year-old Liberty German were killed by members of a pagan Norse religion and white nationalist group known as Odinists.They also requested a hearing to present evidence that the search warrant for Mr. Allen's house was based on faulty probable cause and should be suppressed.

“Members of a pagan Norse religion, called Odinism, hijacked by white nationalists, ritualistically sacrificed Abigail Williams and Liberty German,” the attorneys state said in the filing.

The attorneys claim that two groups of Odinists, one from the Delphi area and one from the Rushville area in southeastern Indiana, were investigated for their involvement in the crimes. They say investigators found multiple ritualistic symbols at the crime scene, including the way Liberty’s body was positioned.

The Carroll County prosecutor’s office didn’t immediately reply to a Monday phone message seeking comment about the defense’s claims.

The two girls were found dead near the Monon High Bridge on Feb. 14, 2017. An autopsy showed that they had been stabbed.

In a search warrant request in March 2017, an FBI agent claimed the girls’ bodies appeared to have been “moved and staged” at the crime scene.

The defense filing claims possible “Odinism signatures," including ritualistic symbols, were left at the crime scene but that investigators abandoned that angle.

Mr. Allen’s attorneys also named several potential suspects who have not been charged in the case.

The girls were killed after a relative dropped them off at a hiking trail near the Monon High Bridge just outside their hometown of Delphi, about 60 miles northwest of Indianapolis. Their bodies were found the next day in a rugged, heavily wooded area near the trail.

Mr. Allen has pleaded not guilty to the charges.

The killings have haunted Delphi, a city of about 3,000 where Mr. Allen lived and worked at a drug store.

Tour at Rice University, calculates that the commercial value of the graphene should more than offset the costs of the process, essentially offering a free source of hydrogen gas.

“This is a solved problem now, to get hydrogen for free—it just has to be scaled up in industry,” Tour says.

“I think it’s a really important advance, it’s very exciting,” says Peter Edwards at the University of Oxford, who has developed a microwave-based method for extracting hydrogen from plastics, and was not involved in the research.

The United States recycles less than 10% of its plastic waste. The remainder, often containing intractable mixtures of different plastics, is incinerated, sent to landfills, or discarded as litter. Although pyrolysis can convert mixed plastic waste into useful hydrocarbons, this energy intensive approach has also raised environmental concerns.

Meanwhile, there is huge demand for hydrogen as a clean-burning fuel and chemical feedstock. Yet more than 95% of global hydrogen production comes from steam-methane reforming, which generates 11 kg of carbon dioxide per kg of H 2 , and using renewable sources of electricity to produce ‘green’ hydrogen relies on expensive metal catalysts such as platinum.

Tour says his process can tackle both of these challenges at once, because plastics are such a rich source of hydrogen atoms. The method relies on flash Joule heating, in which a burst of electrical current superheats samples until they decompose. Tour’s team has previously used this to recycle graphite battery anodes, and to produce graphene from petroleum waste, coal and plastic. Although they didn’t know it at the time, their previous experiments on plastics also released hydrogen, says Tour.

In the latest work, the researchers ground up polyethylene and added small amounts of a conductive additive such as carbon black. Then they loaded the material into a small quartz tube with a hollow electrode that allowed gas to escape. Several jolts of current, each less than 3 s long, heated the sample to about 2800 °C. This converted up to 93% of the hydrogen atoms present in the polymer into hydrogen gas with a purity of 87%. Using mixed plastic waste also produced hydrogen, albeit at somewhat lower yields and purity.

The carbon atoms left behind formed tiny flakes of graphene, several atoms thick, which agglomerated into disordered stacks. Tour says this material is suitable for use as an additive to strengthen composite materials.

The process joins a growing list of methods to mine hydrogen from plastic waste. “It’s a hot topic,” says Taylor Uekert, a circular economy researcher at the National Renewable Energy Laboratory in Colorado.

Uekert previously worked on a photochemical process to extract hydrogen from waste plastic. Another option uses pyrolysis to turn polymers into small hydrocarbons that can be reformed into H2, CO and CO2. In a third approach, Edwards’s microwave-based technique liberates hydrogen directly, leaving carbon behind as nanotubes.

But Tour points out that flash Joule heating requires no catalyst, and uses less energy than other ways to extract hydrogen from waste plastic. If it runs on renewable electricity, it also produces very little CO2. His team calculates that even if the graphene they produce were sold at just $3 per kg—about 5% of its current market price—that income would more than pay for the operational costs.

Still, Uekert says that mechanical recycling tends to be a very energy-efficient way to turn plastic waste into fresh plastic. From an environmental point of view, mining hydrogen from plastic waste might be best applied to waste that is unsuitable for recycling. “You need to be using the heavily contaminated plastic waste that no one wants to buy,” she says. She adds that the team should carry out a more comprehensive life-cycle analysis of how the process would operate in a pilot-scale plant, including the energy used to purify the hydrogen and the toxicity of any materials involved.

The company Tour cofounded, Universal Matter, plans to operate a demonstration plant in Ontario that will produce roughly 1 metric ton of graphene per day, from waste materials and biomass, by the end of this year. Tour expects they will subsequently try to capture hydrogen released from the process, and insists that harnessing the flammable gas will be no more dangerous than steam-methane reforming. “Once the engineers get hold of this, I don’t think this is going to be a problem at all,” he says.

Wife was attacked today by a red pickup truck while she was blowing leaves in front of the house. This was blatant, she said the truck stop right in front of her and made a van behind him wait and after a short time release chemicals and drove away slowly; leaving her feeling frozen with paralysis. She said it was a white person, that was looking forward all the time, and it wasn't a regular landscaping truck although it had a plywood half box utility trailer it was pulling. She said that was just a used as a ploy. She is too emotional now for me to get further information. Being that, I would add that we should consider the possibility of both vehicles being part of the pursuit if the chemical attack was two staged or if the first left water as a ploy, while the van used air born vented or exhaust deployment of chemicals like I witnessed at the beginning of Covid. Boleskine House: Aleister Crowley and Jimmy Page's portal to hell

 

There are many notorious houses worldwide, but one of the most storied is Boleskine House, the former home of occultist Aleister Crowley and Led Zeppelin guitarist Jimmy Page. 2'nd Fire at Boleskin House

 

Firefighters have tackled another blaze at the ruined former home of notorious occultist Aleister Crowley.

 

Boleskine House along, overlooking Loch Ness, was badly damaged by a blaze in 2015 and the ruin was sold earlier this year.

The group said the fires were started deliberately.

 

Crowley, who died in 1947, was infamous in the late 19th Century and early 20th Century for his promotion of the occult.

 

Jimmy Page bought the house because of the Crowley connection, before later selling it.

 

Police and fire service officers have started an investigation into Wednesday's fire.

 

Det Insp Eddie Ross said: "Our inquiries are at an early stage, although our initial assessment is that this fire was started deliberately.

Moscow has “no doubt” about Western complicity in the attack on Sevastopol last Friday, the Foreign Ministry has said

 

Maria Zakharova, the spokeswoman for the Russian Foreign Ministry, has asserted that US and British intelligence agencies supported Kiev during an attack on Sevastopol last Friday. The Ukrainian assault targeted the headquarters of the Russian Black Sea Fleet.

 

Speaking at a weekly briefing on Wednesday, she said there was “no doubt that this attack was planned with the use of Western surveillance assets, NATO satellite equipment, and spy planes and conducted at the direction of and in close coordination with American and British special services.”

 

The Defense Ministry has reported that the missile strike on the fleet headquarters involved several Ukrainian missiles, with Russian air defenses successfully intercepting some of them. British media outlets have also disclosed that Kiev employed Storm Shadow missiles supplied by the UK in the attack, resulting in significant damage to the building.

 

Zakharova described the incident as one of many in which Kiev “targets Russian regions, using missiles and shells supplied by NATO states,” citing several other recent examples. The goals of the Ukrainian government, she said, are “to draw attention away from the Ukrainian military’s failed attempts at conducting a counteroffensive” and to destabilize Russian society.

 

The US and its allies have supplied tens of billions of dollars worth of military hardware to Ukraine to boost its summer charge against Russian defensive lines. The operation, however, has so far produced insignificant territorial gains at the cost of heavy losses in manpower and equipment.

 

Russian Defense Minister Sergey Shoigu estimated on Tuesday that over 17,000 Ukrainian troops were killed in September alone. Western officials and media have acknowledged that the counteroffensive has not turned out as Kiev and its sponsors had hoped.

 

The administration of US President Joe Biden, however, has urged Congress to keep financing Ukraine for the foreseeable future. An appropriation request for over $24 billion in aid is currently floating in the legislature but is opposed by some Republican lawmakers.

 

Senators have proposed a compromise stopgap spending bill to avoid a possible government shutdown next month. The plan involves reducing spending on Ukraine to $6.2 billion. The GOP-controlled House of Representatives would need to approve the draft before it could be voted on and sent to Biden’s desk to be signed into law.

Alex Gino is the author of “Melissa,” a book about a boy named George who believes he is a girl. The book has been banned in four U.S. states and was No. 1 on the American Library Association’s most challenged list from 2018 to 2020, according to Yahoo News.

 

The book was initially published as “George,” but later renamed “Melissa.”

 

BookLooks, a website that rates children’s books, summarized the concerns in the book as follows: “This book contains alternate gender ideologies including transitioning commentary; inexplicit non-sexual nudity; and alternate sexualities.”

 

One part of the book describes how George feels while taking a bath. The passage reads: “She took off her shirt while the tub filled waiting until the last possible moment to take off her pants and underwear. She immersed her body in the warm water and tried not to think about what was between her legs, but there it was, bobbing in front of her,” according to the Des Moines Register.

 

Gino, who identifies as “nonbinary,” told Yahoo News, “Parental rights really anger me, because what about human rights? People who are under 18 are human and if you are keeping information about the world from young people, you are leaving them less prepared to learn how to be in the world.”

 

“If my book is going to get challenged, that to me is a sign that there are more stories that I need to write. And so I’m writing now about queer and trans kids who don’t just exist but who know each other and who have community and who get to thrive. If they’re going to say, ‘Don’t do it,’ then I better not tell myself not to do it. I better do it more,” he added.

 

According to the LGBT Foundation, “Non-binary is used to describe people who feel their gender cannot be defined within the margins of gender binary. Instead, they understand their gender in a way that goes beyond simply identifying as either a man or woman.”

So let’s let the most confused group of people in the world prepare children for the world instead of the people who care about them the most — that will definitely help them.
