"Targeting tumor hypoxia and mitochondrial metabolism with anti-parasitic drugs to improve radiation response in high-grade gliomas"

Abstract

High-grade gliomas (HGGs), including glioblastoma and diffuse intrinsic pontine glioma, are amongst the most fatal brain tumors. These tumors are associated with a dismal prognosis with a median survival of less than 15 months. Radiotherapy has been the mainstay of treatment of HGGs for decades; however, pronounced radioresistance is the major obstacle towards the successful radiotherapy treatment. Herein, tumor hypoxia is identified as a significant contributor to the radioresistance of HGGs as oxygenation is critical for the effectiveness of radiotherapy. Hypoxia plays a fundamental role in the aggressive and resistant phenotype of all solid tumors, including HGGs, by upregulating hypoxia-inducible factors (HIFs) which stimulate vital enzymes responsible for cancer survival under hypoxic stress. Since current attempts to target tumor hypoxia focus on reducing oxygen demand of tumor cells by decreasing oxygen consumption rate (OCR), an attractive strategy to achieve this is by inhibiting mitochondrial oxidative phosphorylation, as it could decrease OCR, and increase oxygenation, and could therefore improve the radiation response in HGGs. This approach would also help in eradicating the radioresistant glioma stem cells (GSCs) as these predominantly rely on mitochondrial metabolism for survival. Here, we highlight the potential for repurposing anti-parasitic drugs to abolish tumor hypoxia and induce apoptosis of GSCs. Current literature provides compelling evidence that these drugs (atovaquone, ivermectin, proguanil, mefloquine, and quinacrine) could be effective against cancers by mechanisms including inhibition of mitochondrial metabolism and tumor hypoxia and inducing DNA damage. Therefore, combining these drugs with radiotherapy could potentially enhance the radiosensitivity of HGGs. The reported efficacy of these agents against glioblastomas and their ability to penetrate the blood-brain barrier provides further support towards promising results and clinical translation of these agents for HGGs treatment.

Anyone else dealing with this from family, their lives have been ruined by Fauci et al and they think its normal.

My vaxxed sister telling my siblings the solution to having a Merry Christmas, test all that comethey are only inviting close family. But why test when they come, when not rnjoy the day and test at night?

My family normally had a big get together with my 9 siblings and children every year for all holidays, now they are all isolating with a few. I’m out of state, but wouldnt go if invited because I got sick last week by meeting two vaxxed clients on my office. I do miss those get togethers though.

An in silico chemical genomics approach is developed to predict drug repositioning (DR) candidates for three types of cancer: glioblastoma, lung cancer, and breast cancer. It is based on a recent large-scale dataset of ~20,000 drug-induced expression profiles in multiple cancer cell lines, which provides i) a global impact of transcriptional perturbation of both known targets and unknown off-targets, and ii) rich information on drug’s mode-of-action. First, the drug-induced expression profile is shown more effective than other information, such as the drug structure or known target, using multiple HTS datasets as unbiased benchmarks. Particularly, the utility of our method was robustly demonstrated in identifying novel DR candidates. Second, we predicted 14 high-scoring DR candidates solely based on expression signatures. EIGHT OF THE FOUTEEN DRUGS SHOWED SIGNIFICANT ANTI-PROLIFERATIVE ACTIVITY AGAINST GLIOBLASTOMA, I.E., (((IVERMECTIN))), trifluridine, astemizole, amlodipine, maprotiline, apomorphine, mometasone, and nortriptyline. Our DR score strongly correlated with that of cell-based experimental results; the top seven DR candidates were positive, corresponding to an approximately 20-fold enrichment compared with conventional HTS. Despite diverse original indications and known targets, the perturbed pathways of active DR candidates show five distinct patterns that form tight clusters together with one or more known cancer drugs, suggesting common transcriptome-level mechanisms of anti-proliferative activity.

In our work, we show that an anthelmintic drug, ivermectin, is active against glioblastoma cells in vitro and in vivo, and also targets angiogenesis. Ivermectin significantly inhibits growth and anchorage-independent colony formation in U87 and T98G glioblastoma cells. It induces apoptosis in these cells through a caspase-dependent manner. Ivermectin significantly suppresses the growth of two independent glioblastoma xenograft mouse models. In addition, ivermectin effectively targets angiogenesis through inhibiting capillary network formation, proliferation and survival in human brain microvascular endothelial cell (HBMEC). Mechanistically, ivermectin decreases mitochondrial respiration, membrane potential, ATP levels and increases mitochondrial superoxide in U87, T98G and HBMEC cells exposed to ivermectin. The inhibitory effects ivermectin are significantly reversed in mitochondria-deficient cells or cells treated with antioxidants, further confirming that ivermectin acts through mitochondrial respiration inhibition and induction of oxidative stress. Importantly, we show that ivermectin suppresses phosphorylation of Akt, mTOR and ribosomal S6 in glioblastoma and HBMEC cells, suggesting its inhibitory role in deactivating Akt/mTOR pathway. Altogether, our work demonstrates that ivermectin is a useful addition to the treatment armamentarium for glioblastoma. Our work also highlights the therapeutic value of targeting mitochondrial metabolism in glioblastoma.

(CNN)Former President Donald Trump was booed by a portion of an audience in Dallas on Sunday when he said he had received a Covid-19 booster shot, according to video of the closed press event that was shared on social media.

The comments by Trump who, despite championing his administration's efforts to develop Covid vaccines, rarely discusses his own vaccination and has largely declined to encourage others to get it came during a stop of his tour with former Fox News host Bill O'Reilly.

According to video tweeted by O'Reilly's "No Spin News," the former Fox News host says, "Both the President and I are vaxxed" and then asks Trump, "Did you get the booster?"

"Yes," Trump says to a smattering of boos in the audience. "Don't, don't, don't, don't, don't," Trump says in the video, seemingly trying to quiet the boos. "That's all right, it's a very tiny group over there."

CNN has reached out to a Trump spokesperson for more details on the former President's decision to get the booster shot.

In a longer video later tweeted by O'Reilly's site, Trump warns supporters that they are "playing right into their hands" when they dismiss the vaccines and don't take credit for them.

Deadly 'Black Fungus' Disease Spotted in Americans With COVID-19

THURSDAY, Dec. 16, 2021 (HealthDay News) It's a phenomenon first identified in India earlier this year: Patients who have or are recovering from COVID-19 who then contract a sometimes deadly fungal infection known as mucormycosis  also known as "black fungus."

Now, the U.S. Centers for Disease Control and Prevention said isolated cases of the disease are hitting COVID patients in the United States.

"During Sept. 17-24, 2021, three clinicians independently notified the Arkansas Department of Health [ADH] of multiple patients with mucormycosis after a recent diagnosis of COVID-19," CDC researchers reported.

The condition is caused by a variety of naturally occurring fungi that are typically harmless, but can trigger illness in folks whose immune systems have been depleted by illness, including COVID-19.

In a statement issued by the Indian Ministry of Health and Family Welfare in May, experts there explained that "people catch mucormycosis by coming in contact with the fungal spores in the environment [soil or decomposing leaves]. It can also develop on the skin after the fungus enters the skin through a cut, scrape, burn or other type of skin trauma.

"Mucormycosis begins to manifest as skin infection in the air pockets located behind our forehead, nose, cheekbones, and in between the eyes and teeth," the Indian agency added. "It then spreads to eyes, lungs and can even spread to the brain. It leads to blackening or discoloration over the nose, blurred or double vision, chest pain, breathing difficulties and coughing of blood."

Once established, "black fungus" disease is tough to treat. As the Indian experts explained, "treatment involves surgically removing all dead and infected tissue. In some patients, this may result in loss of upper jaw or sometimes even the eye. Cure may also involve a 4-6-week course of intravenous antifungal therapy. Since it affects various parts of the body, treatment requires a team of microbiologists, internal medicine specialists, intensivist neurologists, ENT specialists, ophthalmologists, dentists, surgeons and others."

Reporting in the Dec. 17 issue of the CDC journal Morbidity and Mortality Weekly Report, researchers led by CDC epidemiologist Dr. Jeremy Gold said they identified 10 lab-confirmed cases of black fungus illness in patients treated at six Arkansas hospitals between July 12 and Sept. 28, 2021. Nine of the 10 patients lived in the state, all were white, seven were men and the average patient age was 57.

All had tested positive for COVID within the prior two months, and eight of the 10 patients also had diabetes – another noted risk factor for contracting mucormycosis, the researchers noted.

Many cases were severe – four patients showed disease that had spread to the nose and mouth, with three of those patients also having the brain affected. In two cases, the illness attacked the lungs, and in one case the gastrointestinal system was affected, Gold's team said.

None of the patients had been vaccinated against the new coronavirus.

Besides battling mucormycosis, eight of the patients suffered such severe cases of COVID-19 that they required either supplemental oxygen or mechanical ventilation to breathe, the researchers said.

Most patients did not survive their ordeal: "Five patients received surgical treatment to excise mucormycosis-affected tissue," the CDC researchers said, and "six of the 10 patients died during hospitalization or within one week of discharge."

The team noted that the outbreak in black fungus cases in Arkansas coincided with a midsummer statewide surge in COVID-19 cases, driven by the emergence and spread of the Delta variant.

In the absence of COVID-19, mucormycosis is exceedingly rare in Arkansas or other states. However, based on the summer outbreak, the Arkansas Department of Health "coordinated a statewide call on Oct. 11, 2021 to infection preventionists for COVID-19-associated mucormycosis cases," the researchers said.

Dr. Amesh Adalja is senior scholar at the Center for Health Security at Johns Hopkins in Baltimore. He wasn't involved in the new report, but said "it is not surprising that mucormycosis is also able to 'super-infect' COVID-19 patients who have severe immune dysregulation."

As they battle COVID-19, some of these patients may also be receiving medications that suppress their immune systems, such as dexamethasone or tocilizumab, and many will have already suffered lung damage, Adalja pointed out. That leaves them even more vulnerable to fungal infections such as mucormycosis.

Of course, many of the tragedies outlined in the Arkansas report could have been easily avoided, he added.

"The best prevention is to not have a case of severe COVID-19 in the first place, by being vaccinated," Adalja said.
